The Promise of Induced Pluripotent Stem Cells: Basic Research and Clinical grade manufacturing

Pluripotent Stem cells (PSC) research has generated tremendous excitement, and holds promise for the treatment of many uncured diseases. Early stage clinical trials using differentiated induced pluripotent stem cells (iPS) show great potential in cell-based therapeutics. Bridging the gap between research models and clinical applications is the utmost goal to ensure clinically relevant stem cell products.

In the present innovation showcase we will present a research work on SNF5 (also known as INI1 and SAMRCB1) that is one of the core subunits of the SWI/SNF chromatin-remodeling complex. Mutations in this protein might have significant effects on the epigenetic state of human embryonic stem cells and on their phenotype and precise levels of SNF5 are required in order to preserve the pluripotent state of the cells.

Critical part of the translation from research to clinical therapeutics is the generation of iPS cells, expansion, and storage of the cells under current Good Manufacturing Practice (cGMP) conditions. The innovation showcase will address the challenges related to the manufacturing of clinical grade human iPS cells including tissue biopsies, xeno-free culture procedures and raw materials, testing and cryopreservation.

A unique defined pre-coating-free culture platform for isolation and expansion of hMSC

This poster evaluates different combinations of treated plasticware (uncoated) and SF, XF MSC media. Results show that MSC NutriStem® XF (BI) together with Corning® CellBIND® (uncoated cultureware) is a superior platform for the expansion and proliferation of hMSC from a variety of sources. Expansion of pluripotent hPSC in various vitronectin-based culture systems

The poster evaluates long term expansion of hPSC using NutriStem® V9 XF on Vitronectin ACF. Results show that NutriStem® V9 XF medium enables high proliferation rate in long-term culture, while maintaining stable karyotype, high pluripotency marker expression, and preservation of tri-lineage differentiation potential of hPSC.
