Product Overview:
MCDB 131 Medium was originally developed by Knedler and Ham for the support of human microvascular endothelial cell (HMVEC) growth. Initially created through adjustments of MCDB 402, MCDB 131 has been optimized for reduced levels of serum supplementation (Knedler & Ham, 1987). While designed for HMVECs, this medium has also been shown to be effective in definitive endoderm differentiation of human pluripotent stem cells (hPSCs), allowing for the derivation of cell lines that produce epithelial lining of systems such as the intestines, lungs, liver, and pancreas. This medium thus provides an optimally balanced nutritional environment for various mammalian cell types. Most specifically, MCDB 131 has proven to be a critical component in the differentiation of hPSCs into functional pancreatic beta cells through several culture stages producing definitive endoderm, primitive gut tube, pancreatic progenitors, endocrine progenitors, and finally, insulin-producing cells (Wang, et al., 2019).
Starting with xeno-free, serum-free NutriStem® hPSC Medium, Biological Industries USA can provide clean culture components for the generation of healthy iPSCs which can thereafter be differentiated in MCDB 131 for the production of functioning, insulin-producing beta cells. Both NutriStem® hPSC Medium and MCDB 131 are manufactured in a cGMP environment.
