Product Overview
Vitronectin is a secreted glycoprotein that can support cell adhesion through binding to various integrins and proteoglycans. Recombinant vitronectin ACF can function as a chemically-defined matrix component for the attachment of human embryonic and induced pluripotent stem cells in a feeder-free culture system. Recombinant Vitronectin ACF is a 459 amino acid, single-chain, monomeric protein, which migrates at an apparent molecular weight of 75 kDa by SDS-PAGE under reducing conditions. The calculated molecular weight of Vitronectin ACF is 52.2 kDa.

- Vitronectin ACF: easy to use and great for generating hiPS cellsReview by Dr. Christina Warnecke

I stored the reconstituted solution in 60 µl aliquots at -80°C. With one aliquot diluted in 12 ml PBS, you can pre-coat at least two 6-well plates, so 200 µg are sufficient for at least 13 6-well plates. The protocols are provided with the substance. To keep the procedure short, you can also directly add the substance to the cells/medium (which is useful when you have to split the cells urgently e.g. on a weekend), but I routinely pre-coated the plates for 1 h. The fibroblasts were efficiently reprogrammed to stem cells and the stem cells grew and looked very well on Vitronectin. On recombinant laminin 511 E8 fragment, which I also tried for comparison, the stem cells seemed to adhere more tightly than on Vitronectin ACF, but this is rather a disadvantage, since the cells should be passaged with EDTA and mechanical disloging (generating lumps but not single cells). So for routine culture and passaging, I prefer Vitronectin ACF. I already re-ordered it several times. (Posted on 4/20/2020)
